About Cali Kaye:
During my undergraduate studies at Queen’s University, I developed a strong interest in genetics and molecular biology while completing an honours thesis in the diCenzo Lab. I successfully used CRISPR-Cas9 to knock out the antimicrobial resistance gene vim-1, an emerging target of interest in public health research. This project gave me foundational training in techniques such as primer design, molecular cloning, PCR, gel electrophoresis, bacterial transformation, cell culture maintenance, and the Kirby-Bauer assay for antibiotic susceptibility.
As a Master’s student in the Singh Lab at McGill University, I am further developing my scientific and technical competencies. My thesis focuses on CRISPR-Cas9-mediated gene editing in barley. I designed and inserted a CRISPR-Cas9 construct into barley and gained experience with plant genetic transformation, DNA extraction, restriction digestion, and the preparation of samples for both deep sequencing and Sanger sequencing.
In parallel, I expressed and purified recombinant proteins in E. coli, which involved troubleshooting solubility issues and optimizing expression conditions. Through this, I became proficient in SDS-PAGE, Bradford assays, and β-glucanase activity assays. These experiences have strengthened my ability to keep meticulous lab records, independently plan and execute experiments, troubleshoot protocols, and adapt my approach when faced with experimental challenges.
